We have previously found that γH2AX inducibility by 4-chloro-o-toluidine (4-Cl-OT) was stronger than that by o-toluidine (OT) by in cultured human urothelial cells. In this study, we examined the underlying mechanisms of the DNA damage induction by 4-Cl-OT. We found that 4-Cl-OT exposure increased intracellular ROS, which is almost completely attenuated in the presence of NAC, an antioxidant. However γH2AX induction by 4-Cl-OT was not effectively attenuated in the presence of NAC. These results suggested that γH2AX induction by 4-Cl-OT is independent of ROS generation.
