[Purpose] Supersulfide such as hydropersulfide (R-SSH) and R-SSS-R' with a sulfane sulfur, is known to function as a powerful antioxidant. Previously, we have reported that supersulfide exists in biological fluids like human blood and saliva, and the relationship between supersulfide and the biological activities of each component. Since stratum corneum (SC) is constantly exposed to oxidative stress by ultraviolet radiation (UV) in sunlight, supersulfide is possible to play the important role in an oxidative stress defense mechanism. In this study, we examined to detect the supersulfide contained in SC, and to identify some protein hydropersulfides.
[Methods] SC was collected according to the SC tape strip method as previously reported. SC adhering to the tape was solubilized by sonication in boric acid buffer containing 0.1% Triton X-100 on ice. The supersulfide in the solubilized SC was quantified by the elimination method of sulfide from polysulfide (EMSP). The detection of protein hydropersulfides was performed using the protein persulfide detection protocol (ProPerDP).
[Results and Discussion] The results of EMSP showed that supersulfide is present in SC of the skin and oxidative stress by ultraviolet rays could consume the supersulfide. In addition, when we identified protein hydropersulfides by ProPerDP, keratin 10 was identified. We are ongoing to examine the antioxidant mechanism of supersulfide contained in keratin 10 and the relationship between supersulfide and skin diseases using a psoriasis model.
