Fundamentals of Read-across and the Status of its Regulatory Use

Abstract

Reed-across is a kind of alternative methods to animal testing. It is a method to group chemicals with similar chemical structures etc. and estimate the hazardous of untested chemical from the test data of chemicals in the group (analogue chemicals). The scientific evidence for grouping chemicals is important for giving a reliable estimation by read-across. The various elements that can be scientific evidence for grouping are summarized in the OECD guidance document on grouping of chemicals. In particular, the importance of grouping of chemicals based on the mechanism of toxicity is emphasized, and as one of the methods, grouping based on the substructure that causes the toxicity is recommended. In this presentation, fundamentals of read-across will be summarized based on the OECD guidance. In addition, free computational tools for supporting evaluation by read-across (OECD QSAR Toolbox and HESS system) will be introduced.

Read-across enables us to make highly transparent and careful evaluations based on various perspectives. Since such advantages of read-across are suitable for regulatory use, it is widely used in the regulation of chemicals in each country, such as EU REACH, US TSCA and Japan’s CSCL. In this presentation, the comparison of the purpose and scope of the use of read-across in each regulation will be shown.

The OECD is developing case studies on the regulatory use of alternative methods in each country as an activity to promote the regulatory use of alternative methods. These case studies are used for the discussions on the improvement of alternative methods or possibility of the use of the evaluation methods in other countries, etc. The most frequently used alternative method in currently published case studies is read-across. In this presentation, some discussion points of the read-across case studies will be introduced.
