Liquid biopsy using cfDNA methylation and EV-associated miRNA as a toxicity biomarker

Abstract

Recently, it has been reported that cell free DNA (cfDNA) and exosomal RNAs are circulating in the blood. Since there are DNA methylation sites specific to various organs in the body, cfDNA with DNA methylation specific to the target organ would leak into the blood stream when the target organ is damaged. It has been also well known that exosomal RNAs could be used as a cancer biomarker for early diagnosis. Here, we introduce a novel toxicity evaluation method using DNA methylation of cfDNA and exosomal RNA as toxicity biomarkers in mouse blood after administration of various drugs and chemicals. In the blood of mice with liver injury, cfDNA with a liver-specific DNA methylation pattern increased, and liver injury-specific exosomal RNA was also induced.

The liquid biopsy using cfDNA methylation and EV-associated miRNAs as toxicity biomarkers would accelerate a rapid evaluation of chemical substances and drugs in Nonclinical Safety Evaluation.
