The bar gene was introduced into the suspension culture of creeping bentgrass (Agrostis palustris Huds.) ‘Chiba green B-1’, a “Brown patch” -resitant cultivar, co-transformed with the gus or cry I A (c) FM gene by particle bombardment. 16 transgenic plants were obtained after the selecion on the medium containing bialaplios.
PCR and PCR-Southern hybridization analyses showed that all the bialaphos resistant regenerants tested carried the bar gene . Some plants appeared to be cointegrated with the gus or cry IA (c) FM gene.
Expression of the bar gene was detected by the determination of ammonia content in leaf extract after spraying with 0.5 % (Vol/Vol) HERBIETM. Ammonia contents of transgenic plants were very low as compared with untransformed control. Expression of the cry IA (c) FM gene is under investigation.
