Optimization of callus induction and plant regeneration from seed explants of Zoysia species

Abstract

The effect of genotype, basal medium and silver nitrate (AgNO3) on callus induction and plant regeneration from seed ofZoysiaspecies has been investigated. Surface-sterilized seeds were cultured in LS medium supplemented with 5 mg/l 2, 4-D, 0.1 mg/l BA, 4 mg/l thiamine-HCl, 100 mg/l α-ketoglutaric acid, 3% sucrose and 0.2% gelrite. The friable calli were induced from scutellum of the seeds, and were soft and translucent. On the other hand, the compact callus which were secondary induced from the friable calli, were nodular and yellowish-white in color. The compact calli were transferred to the regeneration medium, LS medium supplemented with 1 mg/l BA, 3% maltose and 0.5% gelrite. The frequency of compact callus induction and plant regeneration were different between genotypes. When comparing LS medium with Ogawa medium, no difference was observed between compact callus induction and plant regeneration. The medium containing 5 and 10 mg/l AgNO3significantly increased the frequency of compact callus induction and shoot formation. As a result of the histological observation, about 2 weeks after transfer into the regeneration medium, rounded structures were found on the surface of the growing compact calli, with supplemental two leaf primordia emerging on the callus surface.
