Dietary vanillic acid prevents postmenopausal symptoms in ovariectomized mice

Abstract

Risk of developing metabolic diseases such as osteoporosis, obesity, and diabetes increases in postmenopausal women. Thus, chemoprophylaxis of postmenopausal symptoms by daily diet is important. Vanillic acid (VA) is a phenol compound found in the herbal medicine ”Toki” (Angelica acutiloba) and in the mycelium of the shiitake mushroom Lentinula edodes. The compound showed no affinity to estrogen receptors and may exert preventive actions of disease. The present study demonstrated the metabolic effects of vanillic acid on ovariectomized (OVX) mice. Ten-week-old OVX mice were fed a VA-containing diet (100 mg/kg body weight/day) for 10 weeks. Dietary VA significantly prevented femoral bone mineral density (BMD) loss corresponding with decreased deoxypyridinoline (DPD) in urine and tartrate-resistant acid phosphatase (TRAP) activity in serum. Dietary VA also decreased the incremental in glucose levels and white adipose tissue (WAT) weights as compared with those in the OVX control group. Furthermore, uterine enlargement was not observed owing to these beneficial effects. Our findings collectively suggest that VA has the potential to retar postmenopausal symptoms in OVX mice without estrogenic action.
