Rate of oxidation by American cockroach muscle mitochondria increased and respiratory control was high level when the mitochondria were isolated with a medium containing Trisbuffer, bovine albumin and cysteine. Rates of pyruvate, α-glycerophosphate and succinate oxidation were stimulated and a value of respiratory control was higher than 1 only in the case of pyruvate.Various inhibitors and uncouplers inhibited mitochondrial respriation of roach muscle prepared with medium containing sucrose (250 μM), EDTA (1 mM), Tris-buffer (10 mM), bovine albumin (0.1%) and cysteine (0.05%). Nitrophenols, chlorophenols and a cyanophenol showed uncoupling functions. The difference on the degree of inhibition to rat liver mitochondria between p-nitrophenol and 3-methyl-4-nitrophenol, metablites of methyl parathion and Sumithion respectively, was small. Lindane did not uncouple the mitochondrial respriation.
