Two-dimensional electrophoresis is a powerful separation method for proteins and/or peptides mixture. Recently the Model TEP-1 was developed, which allows fully automated transfer from the first-dimensional IEF gel to the second-dimensional SDS-PAGE gel under a microprocessor control. The present paper describes the instrumentation and applications of the TEP-1 for peptides and proteins. The TEP-1 can be also used for rapid sample preparation in microsequence analysis. The internal sequence can be identified by site-specific fragmentations of various proteins.
