Pharmacological effects of SF-837, a new macrolide antibiotic, were studied and following results were obtained.
1) In urethane (1g/kg, s. c.) anesthetized rabbit, 20mg/kg (i. p.) SF-837 showed depressor effect, but no effect on respiration and ECG. When 100mg/kg SF-837 was administered orally, almost the same effects were observed. These cardiovascular effects were retained after the administration of atropine and diphenhydramine, vagotomy,and spinal resection.
2) The movements of isolated rat uterus and rabbit intestine by MAGNUS' method were inhibited by 0.01% SF-837 and 0.1% SF-837 caused relaxation on isolated guinea-pig trachea muscle in MAGNUS' method.These effects on isolated smooth muscle were antagonized by ACh and BaCl2.
3) No effect was observed by the administration of 0.2% SF-837 on the plate of diaphragm nervemuscle preparation in rat.
4) For the examination of formation of antibody in rabbit, MATSUHACHI's method was used. SF-873 was administered bcutaneously twice a week 50mg/animal for 4 weeks and the blood was taken on 8th week. SF-837 did not form any antibody in rabbit.
5) SF-837 at 10% caused a slight irritation to the subcutaneous tissue and muscle of rabbit and 500mg/kg caused slight irritation to the gastric mucosa of rat.
6) No effect was observed on body temperature when 200mg/kg of SF-837 was administered orally.
