Abstract

Hitherto histamine, serotonin and acetylcholine have been the chemical mediators in allergic and anaphylactic reactions. From these points of view many anti-a llergy drugs have been synthesized. However, as the author described in the 1st report, bradykinin might be one of chemical mediators in allergic or inflammatory reactions. On the other hand, only aspirin and phenylbutazone have been reported as antibradykinin drugs and few works have been done on anti-bradykinin drugs. Recently Homochlorcyclidine(SA-97) was synthesized by Abbott Laboratory as anti-allergy drug having anti-bradykinin activity. The author studied on the anti-allergic and anti-bradykinin activities of many kinds of anti-allergy drugs and the following results were obtained. 1)The pretreatment of SA-97(Homochlorcyclidine) at the concentration of 0.52mg/kg showed 80% protective effect against anaphylactic shock in the guinea pig. 2)Anti-bradykinin activities of 12 kinds of anti-allergy drugs were tested in vitro by Schutz-Dale technique. The intestinal contraction by 0.1mcg/ml of bradykinin was completely inhibited by SA-97 at the concentration as low as 0.1mcg/ml. Among the tested anti-allergy drugs, SA-97, diphenylpyraline and cyproheptadine have powerful anti-bradykinin activities. 3)It was demonstrated that these three anti-allergy drugs have anti-histamine, anti-serotonin, and anti-acetylcholine activities as well as anti-bradykinin activity and also anti-anaphylactic acti-vity. It was also found that anti-allergy drugs other than these drugs such as pyribenzamine have only slight anti-bradykinin activity.
