Study on Effects of Prostaglandins upon Ciliary Activity of Respiratory Epithelium in Experimental Bonchial Asthma : Part 1. In Vitro Experiment

Abstract
The change of ciliary movement of the respiratory epithelium due to the effect of prostaglandin(PG)E_1, F_<2α>, histamine, acetylcholine and the "Maiko" allergen was investigated and compared among nonsensitized and sensitized guinea pigs with this allergen. The frequency of ciliary beats was estimated by cinematographic recording method using by the phase difference microscope. The following results were obtained. 1) The ciliary movement in the solutions of 10^<-8> to 10^<-5> g/ml PG E_1 decreased for 20 minutes, increased for 30 minutes and then decreased again. 2) The ciliary activity in the solutions of 10^<-7> and 10^<-6> PG F_<2α> showed no change but tended to decrease in the solutions 10^<-5> and 10^<-4>. 3) In sensitized guinea pigs, the depressed ciliary activity by the causative antigen was significantly improved by the solutions 10^<-8> and 10^<-5> PG E_1 but was not influenced in the solution of PG F_<2α>. 4) Simultaneous application of histamine and PG E_1 or PG F_<2α> showed no peculiar difference among them on ciliary movement. 5) Highly diluted acetylcholine(10^<-8>-10^<-4>)solution did not suppress the ciliary movement except the 10^<-2> solution which showed some effect of suppression. Simultaneous application of 10^<-8> acetylcholine and 10^<-6> PG F_<2α> solutions suppressed significantly the ciliary movement. Thus synergor effect between acetylcholine and PG F_<2α> was demonstrated.
