Abstract
A Japanese female with DNA autosensitivity, manifesting chronic bleeding skin ulcers and spontaneous painful ecchymoses of the extremities, nosebleed, hematemesis, rectal and uterine bleeding, hematuria, aphthae, and bronchial asthma is reported. Rejection of autologous skin grafts on the bleeding ulcers occurred on seven occasions over a 12-year period. Painful, tender erythematous swelling and ecchymoses identical with spontaneous ecchymoses were produced by intradermal injections of lysed autologous leukocytes and calf thymus DNA. In vitro incubation of either substance with DNase or chloroquine sulfate abolished these skin reactions. Passive cutaneous anaphylaxis (PCA) test using calf thymus DNA, prodcuced a hemorrhagic cutaneous reaction with blueing. Chloroquine sulfate therapy resulted in the complete suppression of bleeding from all different sites and of the asthma attacks.
