The antibacterial activity of a new antibiotic substance gentamicin against Gram-negative bacilli, Bacil, pyocyaneus, Proteus vulgaris, Esch. coli, and Staphylococci, was excellent. Its blood level following an intramuscular injection of 40 mg/kg was about 5μg/ml. Urinary excretion was good, but transfer into the bile was no good.
The blood level was indeterminable following its oral administration. Given by this route, the drug was very little excreted in the urine.
Used on 4 cases of Bacil. pyopyaneus infection, the drug proved good. But, BUN rose in one case. Clinical findings suggestive of vestibular dysfunction were obtained in another case.
Liver functional tests showed no particular abnormality in the clinical cases. But, experimental studies observed abnormal GOT and GPT among the animals. Histologically, there was nothing abnormal observed by optical microscopy. Electron microscopic examination found some abnormalities. This finding by electron microscopy together with the results of the liver functional tests has made us presume that the drug has the possibility of producing liver disturbance.
