Gantamicin sulfate (GM), a broad spectrum antibiotic, was examined for blood level, urinary excretion, antibacterial activity, clinical effectiveness, and side effects.
Following a single intramuscular injection of 40 mg GM, t he blood level at 30 minutes was 7. 0 mcg/ ml; even at 8 hours after the administration, the blood level was 0. 31 mcg/ml. Following the a dministration of the same dose as above the total urinary excretion for 24 hours was 31. 0 mg, which was 77% of the administered dose.
Sensitivity to GM was examined in 95 strains of Bacillus pyocyaneus, 54 of Esch. con, 37 of Proteus vulgaris, 7 of Klebsiella, and 67 of Staph. aureus. All the other strains than 4 strains of Bacillus pyocyaneus had their growth inhibited by not more than 2. 5 mcg/m1 of GM.
GM was effective in 12 of 17 cases of surgical infections and ineffectiv e in the remaining 5 cases.
Examination of blood picture, liver functions, and renal functions in the cases given GM foun d tra. nsitional renal hypofunction in one of them. This hypofunction was restored to normal following. the termination of the drug administration.
