Abstract
We experimentally and clinically investigated cefpirome (CPR) a newly developed injectable antibiotic.
The serum level was studied in dehydrated and over-hydrated rabbits after a bolus i.v. injection of 40mg/kg of CPR. The serum level was higher in dehydrated than in overhydrated rabbit. CPR was given intraperitoneal administration at 40mg/kg to rat. The concentration of CPR in rat kidney and testis tissue was not detectable at 8 h, nor inepididymis tissue at 6 h.
CPR was given i. v. at 1g to patients who were about to undergo archiectomy before 20-60 min. We could not estimate the time-dependency of the concentration of CPR in human testis because of a few samples.
In a clinical study, 1g of CPR was given by i. v. drip infusion for several days to two patients with acute simple pyelonephritis and nine patients with chronic complicated urinary tract infection.
Efficacy was moderate in five patients and poor in three according to the criteria of the Japanese UTI Committee. The efficacy rate was 62.5%.
No adverse reactions or objective and subjective signs occurred, nor were abnormal clinical laboratory data due to CPR observed.
