Acute nephrotoxicity studies of biapenem in rabbits or rats

Abstract

The present studies were conducted to evaluate the potential nephrotoxicities of biapenem (BIPM), anew carbapenem antibiotic, when intravenously administered to normal rabbits or nephropathy rats.The results obtained are summarized as follows:
1. Single dose nephrotoxicity study in rabbits: Remarkable nephrotoxic changes were observed inall of the rabbits given CER, but no remarkable changes in the kidney were observed in all of the animals given BIPM or IPM/CS.
2. Single intravenous renal toxicity studies of BIPM co-administered with furosemide and glycerin in rats: The renal toxicity was enhanced by co-administration of CER. On the other hand, BIPM and the other reference compounds showed no modulating effects on the renal toxicity in these test systems.
3. Single dose nephrotoxicity study in experimental nephropathy rats: Animals in the vehicle control group preadministered with gentamicin showed moderate changes in the functional indices. In the CER group, changes of the parameter mentioned were severer than those in the vehicle control group, and it was indicated that CER might have nephrotoxic potential in this nephropathy model. On the other hand, BIPM and the other reference compounds showed no drug related nephrotic potential effects in the nephropathy rats to be compared with the animals in the vehicle control group.
