Usefulness of CI-581 [2- (O-Chlorophenyl) -2-Methyl Aminocyclohexanone Hydrochloride] as a Short Acting Anesthetic Drug for Cynomolgus Monkey (Macaca irus)

Abstract
Ten cynomolgus monkeys of 3-5 years of age weighing from 2.30 to 3.33 kg were administered intramuscularly with CI-581. For 5 consecutive days, the animals were injected with the drug at dose level of 2.5 mg per kg of body weight on the 1st day, 5 mg/kg on the 2 nd day, 10 mg/kg on the 3 rd day, 20 mg/kg on the 4 th day and 40 mg/kg on the last day.
General conditions of the animals were observed, and both the induction and duration times of narcosis were measured. Rectal temperature was taken just before and 30-40 min. after the administration.
In all of the monkeys, the process of induction and duration of narcotic effect was remarkably smooth. The monkeys recovered from the narcosis safely and smoothly.
Table 1. summarized the induction time, the duration time and the decrease of rectal temperature. As shown in the table, the larger the dose used, the shorter the induction time and the longer the duration time.
The present results indicate that CI-581 is a practically useful anesthetic drug for cynomolgus monkey when administered intramuscularly at the dose level of 10-20 mg/kg.
