There have been scarcely any reports regarding the habitforming property of thebaine, a kind of opium alkaloid, and opinions differed as to whether or not dehydrooxycodeinone, a thebaine derivative, has the same action. The writer previously obtained affirmative results of the presence of the said property in thebaine by experiments on dogs and rabbits. He proceeded with the experiments with thebaine and dehydrooxycodeinone on mice to ascertain the existence of the property through fatal doses and the duration period of the tail-raising reaction. Results: 1. The fatal dose of thebaine-HCl for normal mice was 0, 3 mg. per 10 g. body weight, while that for mice which had been consecutively injected with thebaine for 20 days, was 0, 38 mg. 2. In normal animals as well as in those accustomed to the substance, the relation between the amount of injection of thebaine-HCl (mg.) per 20 g. (M) and the duration period of the tail-raising reaction (minute) (T) could be represented by the same mathematical formula, T=K MP. The difference between them was in K and P only, i. e. T=147, 9 M1, 933; the duration period was greatly shortened. 3. The fatal dose of dehydrooxcodeinone-HCl for normal mice is 2, 0 mg per 10 g. but in those which has consecutive injections of the substance for 20 days, it was 2, 9 mg. 4. The relation between the amount of dehydrooxycodeinone injected and the duration period of the tail-raising reaction in either the former or the latter animals is the same as that shown in the case of thebaine. The duration period of the reaction greatly shortened, being T=291, 74 M0, 65 in the latter group. 5. The above facts definitely prove that both thebaine and its derivative, dehydrooxycodeinoue, possess habit-forming properties. [cf. original (Japanese) P. 175.]
