The observations reported here are concerned with the microbiological studies on the cultivable acid-fast organisms from the leprosy materials.
Most materials were repeatedly washed in sterile saline solution and aqueous solution containing 8.0% of NaOH, and inoculated onto an egg-yolk medium and incubated at 33°C for 3-5 weeks.The colony of cultivated organisms was grey or orange-yellow.
The isolation frequency of cultivated acid-fast organisms from materials was 0%, 17.4%, 25.0%, 28.6% and 90.0% in case of ear-lobe leproma, subcutaneous leproma, nasal mucosa, nasal crusta and nasal washings respectively.
The biological properties of the organisms were not so much different from that of the atypical mycobacteria. However, the organisms showed remarkable morphological changes during the cultivation, especially in their elongation, cytoplasmic granulation and other phenomena, which were not common with other mycobacteria.
