A Study on Saline Extractable Acid Mucopolysaccharide
-Lipoprotein Complexes of Human Aortas in the Process of Atherosclerosis

Human uninvolved intimas and fibrous plaque tissues in aortas which were collected at autopsy within 5h of death in 30 subjects from 50 to 65 years old were shaked gently or homogenized with 0.15M sodium chloride, pH 7.4, to separate acid mucopolysaccharide (AMPS)-lipoprotein complexes.
Ultracentrifugation was performed on the extracts. Five ml of the top layers floated by ultracentrifugation at 40.000rpm for 20h were passed through a column (1.8×70cm) of Bio-Gel A-50m, 100-200 mesh.
The pattern of the absorbance at 280mμ were separated into three fractions (Fr-I, Fr-II and Fr-III). The molecular weight is the order of Fr-I>Fr-II>Fr-III.
1) AMPS, protein and cholesterol were detected in Fr-I and Fr-II which were separated by Bio-Gel A-50m column chromatography.
2) The amounts of protein and lipids in Fr-I are obviously higher in plaque tissues than those in uninvolved intimal tissues. The ratio of lipids to protein in Fr-I were higher in plaque tissues, also.
3) Almost all protein in serum eluted as a large peak from Fr-II to Fr-III. Lipids could be estimated in Fr-II and Fr-III, but no lipid in serum was detected in Fr-I.
4) In arterial walls, there are at least two kinds of saline extractable substances: one is extracted by gentle shaking, the other is not extracted by the same procedure.
5) Apo B was confirmed by the method of immunodiffusion to anti-apo B in Fr-II which was obtained from extracts of uninvolved intimal tissues and plaque tissues by gentle shaking. Therefore, it is considered that Fr-II separated by gentle shaking is AMPS-lipoprotein complexes with apo B. And also, Fr-I is AMPS-lipoprotein complexes without apo B.
6) Quantitative and qualitative differences of AMPS-lipoprotein complexes were found between uninvolved intimal tissues and fibrous plaque tissues.
It might be considered in the process of atherosclerosis that AMPS-lipoprotein complexes will play important roles on the accumulation of lipids in aortic walls.
