Preparation of Monoclonal Anti-human PDGF Antibodies Using Synthetic Peptide as Immunogen

Abstract

Two mouse monoclonal antibodies against human PDGF, PGF 007 (IgG₁) and PGF 033 (IgM), were prepared with immunization with a chemically synthesized peptide corresponding to positions 73 to 97 of human PDGF-B chain. In immunoblotting, these antibodies specifically reacted with purified human PDGF. By use of the synthetic peptide immobilized on microplates and these monoclonal antibodies, ELISA's for human PDGF were prepared. In these ELISA's, both antibodies were able to react with human PDGF even in the presence of human plasma.
