Abstract
The distribution of antimicrobial agents in infected tissue would be essential for their application to the sites of infection. Taking accounts of the viewpoint, we had reported the distribution of 2 nalidixic acid analogs in the tissues which are related to otorhinolaryngology. We investigated here the distribution of nalidixic acid analog DL-8280. This agent was supplied by Daiichi Seiyaku Co., Ltd. Autoradiography was performed 30 minutes after intravenous administration of 14C-DL-8280 in 100 μCi/20mg/kg to JAW rabbit. Such care was taken in filling the carboxymethylcellulose paste into paranasal cavity, nasal cavity, oral cavity and external ear canal not to damage mechanically. Horizontal frozen sections parallel to mandibular basis were cut in 50cm thickness in a cryostadt. Blackening of 14C-DL-8280 was most significant in soft palate and its expanse which was apparently the same gland-like tissues as soft palate when stained which hematoxyline and eosin. Significant radioactivity was recognized in pharynx, the cartilage of external ear canal and muscle. The levels were low in septal cartilage, concha nasalis and cochlea. Blackening was scarcely seen in brain. Macro-autoradiography can be one of the useful means for the evaluation of chemotherapeutic agents for possible clinical application.
