In order to establish an ideal cisplatin (CDDP)-peplomycin (PEP) combined chemotherapy based on pharmacokinetics, we developed a regimen which showed the most potent anticancer activity in fundamental investigations with the use of flow cytometry. The results were described in our first to third reports.
In the present study, we administered CDDP and PEP intraarterially to a patient with left maxillary cancer. Histopathological examination and CT revealed very favorable results, and the efficacy of the regimen was confirmed. The kinetics of cancer cells during this combined chemotherapy were studied in detail by double staining with propidium iodide and fluorescein isothiocyanate after the intraarterial injection of bromodeoxyuridine, and the viability of the cells was determined with fluorescein diacetate. The results suggest that the S phases blocked 24 hours after the intraarterial injection of cisplatin, and the S and G2M phases are blocked 24 hours after the intraarterial injection of peplomycin. These results were correlated with the data of the fundamental investigations reported previously. Changes in viability correlated were with the histopathological findings.
We believe that application of this method will be helpful in elucidating the mechanism of action of anticancer drugs and concomitant therapy against cancer cells.
