Application of Analysis for Typical Steroids in Ointment

Abstract

Typical steroids in ointment were studied by thin layer chromatography (TLC) using a cyanopropyl-chemically bonded stationary phase, prepared from pre-coated silica gel plate treated with toluene solution containing 3-cyanopropyltrichlorosilane (CN). TLC plate with CN was applied to the evaluation of typical steroids in ointment samples. The CN treated plate was developed in a chromatographic chamber at room temperature with chloroformmethanol (95: 5 v/v) to a distance of 8cm from the origin.
After drying, the plate was sprayed with 10% sulfuric acid solution, and heated. Steroid spots on the chromatogram were measured with a TLC densitometer equipped with a dual-wave length TLC scanner at λ_EX 380nm, λ_EM 550nm.
