The influence of thromboxane A2 synthetase inhibitors (OKY-046: (E)-3-[4-(1-imidazolylmethyl) phenyl]-2-propenoic acid hydrochloride monohydrate, OKY-1580: (E)-3-[4-(3-pyridylmethyl) phenyl]-2-methyl-2-propenoic acid) on the aortic intimal thickening was examined. Male New Zealand white rabbits were fed standard food in control group and experimental food that contained OKY-046 or OKY-1580 in standard food (300mg/100g) in experimental groups. In the Experiment I, each rabbits were fed 100mg of experimental foods per day for 31 days. On the third day, intimal injury was mechanically induced by inserting polyethylene tubing into the aortas. In the Experiment II, each rabbits were fed experimental food containing OKY-046 ad libitum for 10 days. Intimal injury was induced on the third day. The animals were sacrificed after injection of Evans blue dye and 2, 000u of heparin. The injured areas were embedded in Epon or Resin and cut 2μ thick. Then intimal thickness was measured.
In both Experiments, there was no significant difference in the intimal thickening after intimal injury for experimental and control rabbits. The plasma level of the chemicals varied and effective level were not maintained all the time in blood. However, it is also considered that the degree of intimal thickening after mechanical injury may not be proportional to the amount of aggregated platelets and platelet derived growth factor. The intimal thickening might be influenced also by other factors not yet defined.
