Fish plays an important role for human life and ecosystem. Pollutants are often released into the aquatic environment made changing of environment. Transient receptor potential (TRP) protein can sense the changes of the environment factors, and in the case of fish, TRP proteins are conceivable sensor of fish to adapt to the environment changing. Now aday, the zebrafish (Danio rerio) has become more important to scientific research as to sharing major organs and tissues as humans. To date, mammalian TRPM2 is well characterized, however the functional propertises and role of zebrafish TRPM2 still lacking. In this research, we show the characterisation and functional role of zebrafish TRPM2 (zTRPM2) heterologuosly expressed in HEK293T cell.
The result showed that cDNA of zTRPM2 has an open reading frame encoding a protein of 1470 amino acids , it expressed in brad range of organs such as brain, spleen and so on. It is also expressed in immune system such as macrophage and neutrophil, in sensoring nervous system such as motoneurons. Interestingly, the functional analysis using calcium imaging revealed that zTRPM2 is activated by pollutants, such as hydrogen peroxide (H2O2), nitrite (NO2-) and NO (SNAP), however, temperature, pH condition and hypoxia is not effect to zTRPM2 channel. Furthermore, the activation of zTRPM2 was inhibited by mammalian TRPM2 inhibitor econazole and clotrimazole. The results suggest a functional role of zTRPM2 in the detection of oxidants that are known as pollutants and zTRPM2 inhibitors may assists in protecting fish from the polluted environments.
