Abstract

In preliminary study where dog femoral arteries were substitued with Gore-Tex (inner diameter, 3.0mm), all grafts were occluded within six weeks after surgery. To obtain better results, a large dose of platlet aggregation inhibitor (Aspirin, Ticlopidine) or a now under-development antithrombin agent (MD-805) was administered before and after surgery. Almost all grafts were found to be patent at antopsy 4-6 weeks after surgery, but their luminal surfaces were not completly endothelial-lined. To promote the formation of endothelial-lined nonthrombogenic luminal surface, autologous endothelial cell seeding of Vascular-D graft (inner diameter 4mm) was performed. In six dogs, both carotid arteries were resected and one was substituted with Vascular-D graft and the other with endothelial cell seeded Vascular-D graft under the adminisration of MD-805. At the sutopsy 4-6 weeks after surgery, seeded grafts were found to have completly endothelial-lined luminal surfaces.
