Extracorporeal circulation with the inhibition of thromboxane A₂ synthetase

Abstract

A specific thromboxane A₂ (TxA₂) blockade OKY-046 (OKY) was utilized as an anticoagulant for hemodialysis with mongrel dogs. About 300μg/kg/min of OKY clearly prevented blood coagulation in blood circuit, while small dose or very high dose of OKY failed to prevant the clotting. In the latter case, the inhibition of cyclooxygenase by OKY had contributed to that result. Reduction of platelet counts was significantly suppressed during hemodialysis with OKY compared to that with heparin. These results indicate that extracorporeal circulation can be performed by the inhibition of intrinsic platelet functions.
