Burkitt's lymphoma Raji cells were mutanized with ethylmethanesulfonate (EMS) and selected in the presence of 6-thioguanine (6-TG). The most rapidly growing 6-TG-resistant clone (NIR10-2 cells) was obtained. The characteristics of the NIR10-2 cells were as follows:
1 The NIR10-2 cells were deficient in the hypoxanthine-guanine phosphoribosyl transferase (HGPRT) activity, because this cell line was sensitive to HAT selective medium.
2 Enzyme-linked immunosorbent assay (ELISA) found that the NIR10-2 cells did not produce human Ig in the culture medium.
3 The NIR10-2 cells grew well in the new culture medium, GIT.
These results suggest that the NIR10-2 cells are suitable for human-human hybridoma production.
