A Simple Assay of Serum Lipid Hydroperoxides

Abstract
For the determination of lipid hydroperoxides, we previously reported a method that involved the reaction with 10-(N-methylcarbamoyl)-3, 7-(dimethylamino)-phenothiazine under catalysis of hemoglobin and measurement of its product methylene blue. In an attempt to apply this method directly to serum, we found that dissociation of lipid hydroperoxides from serum proteins can be achieved by lipoprotein lipase, and the interfering substances, holotransferrin and ascorbic acid, can be eliminated by adding triethylenetetraminehexaacetic acid and ascorbate oxidase. Pretreatment with these ingredients enabled us to perform a simple assay of serum lipid hydroperoxides.
