Introduction: A protein analysis using mass spectrometry revealed that regenerating gene (REG) 1alpha appears to be related to the pathogenesis of RA. Therefore, the present study was conducted to examine the mechanism of REG 1alpha in RA disease progression. Methods: REG 1alpha serum concentration in RA patients (and normal controls) and the effects of REG 1alpha on apoptosis and proliferation of synovial fibroblasts were evaluated. Results: The serum concentrations of REG 1alpha in RA patients were higher than in normal controls. In addition, TNF-alpha upregulated REG 1alpha production from the synovial fibroblasts MH7A cell line. Furthermore, exogenous REG 1alpha protein inhibited MH7A apoptosis and promoted the cell proliferation of MH7A cells. Conclusion: The present study suggested that aberrant REG 1alpha production has a central role in the increased cell growth of synovial fibroblasts resulting in pannus formation in RA.
