The study of the effects of benamocaine on healthy male volunteers in the age range of 27 to 49 years was made by anatherapeusis, starting with a low dose.
Electroencephalogram, electrocardiogram, respirations, blood pressure, GSR, dermal blood flow, bowel sounds, and body temperature were recorded on polygraph, and the complaints by, and the condition of the subjects were observed and recorded during the period from before administration of benamocaine till two to four hours after the administration.
When orally administered at a single oral dose, 5mg or more of benamocaine had an anesthetizing effect on the gastric mucosa but doses up to 80mg had no side effects, with the exception of one subject in whom auriculoventricular arrhythmia, which appeared psychogenic in nature, was observed.
