The analysis of the Asp·Asn·Glu·Gln contents of the calli inoculated and the regenerated rice plantlets revealed that the bigger the shoot-parts of regenerated plantlets (mg in dry weight) were, the higher the amino acid contents (nmol/mg) were, but the bigger the calli-parts of regenerated plantlets were, the lower the amino acid contents were. The amino acid contents of inoculated calli (before regeneration culture) were not influenced by the size of inoculated calli.
These above mentioned results suggested that these amino acids might promote the regeneration frequency of plantlets by the method of two-step liquid regeneration culture. As the results of addition test of these amino acids in the 2nd regeneration liquid medium. Asp, Asn, Glu and Gln promoted clearly plantlets regeneration.
