We reported practical procedures and its applications of electrophoretic transfer named western blotting from polyacrylamide gels into nitrocellulose membranes in SDS-polyacrylamide gel electrophoresis (SDS-PAGE) . The nitrocellulose membrane was stained by the use of immunoperoxidase method. So we could analyze the antigens and antibodies simultaneously. In this paper, we described the details of western blotting and its practical procedures. In addition we mentioned about two examples of research in our laboratory, that is, immunological analysis in Ascaris suum and myoglobin in thymus of myasthenia gravis. As the results, the combination with immunoperoxidase method and western blotting is a very sensitive method for detecting antigens and antibodies, and supposed to be a powerful method for analyizing immune manifestations caused by various diseases.
