A SURVEY OF THE HISTOCHEMICAL TECHNIQUE FOR THE LOCALIZATION OF CHOLINESTERASES

Abstract

Histochemical technique for the localization of cholinesterase by Koelle has been modified and improved.
For the specific ChE determination 20% and for the non-specific ChE 24% Na₂SO₄ were used as storage solutions. Except Mal (NaH maleate: 1.9g, N·NaOH: 10.4ml., q.s 100ml., pH12.6), the same reagents for the incubation solution were used as Koelle's. Optimal pH of the incubation solution for the determination of specific ChE was 6.5-6.7 and for the non-specific ChE was 7.7-7.8. MgCl₂ in the incubation media for the non-specific ChE determination was excluded since magnesium ions inhibited non-specific ChE competitively while they activated specific ChE.
