HISTOCHEMICAL STUDIES ON SH- AND SS-GROUPS, NUCLEIC ACIDS AND SUCCINIC DEHYDROGENASE IN THE HEALING OF EXPERIMENTALLY INDUCED DERMAL TRAUMA

Abstract
Stripping of the horny layer and the epidermis was given on the skin of both rabbits and human beings and skin samples were collected with certain definite intervals for a period of 30 days in order to investigate the changes produced in the contents of SH-group, SS-group, nucleic acids aed succinic dehydrogenase employing the direct (d-) and indirect (i-) methods of nitroprusside reaction (N. P.), DDD method (Barrnett-Seligman), Bennett method, nucleic acid staining and neotetrazolium method (NT) . Stated in the following are the results obtained thereof :
1. Processes of the traumatic healing were divided into the following 6 stages by the characteristic changes observed on the contents of the substances stated above.
Stage I : For 3 hours after stripping (so-called shock stage), various staining reactions remain either unchanged or weakened.
Stage II : From 3 hours after stripping to 12 hours (hypertrophy stage of the basal and the prickle cells), sudden increase in SH-group and the increase in ribonucleic acid and succinic dehydrogenase in the latter half of this stage by d-N. P, method.
Stage III : From 12 hours after stripping to 5-7 days (stage of cell division), SH-group and ribonucleic acid show the strongest colour reaction by d-N. P, method.
Stage IV : From 5-7 days after stripping to 20 days (stage beginning the formation of horny layer), SH-group, succinic acid dehydrogenase and desoxyribonucleic acid show the strongest colour reaction by Bennett method, and SH-group and SS-group show the strongest colour reaction by DDD method in the latter half of this stage.
Stage V : From 20 days after stripping to 30 days (stage of the formation of horny layer and the proliferation of epidermis), SH-group by i-N. P, method and SS-group by DDD method show the strongest colour reaction.
Stage VI : Later than 30 days after stripping (convalescent stage), all colour reactions are restored to normal.
2. Accordingly, it is considered that the increase in SH-group by d-N. P. method indicates the preventive reaction and the same by i-N. P. and DDD methods is related to the cornification, while the increase in SS-group by DDD method suggests the formation of horny layer.
