EFFECT OF DICLOFENAC SODIUM ON BOVINE LENS EPITHELIAL CELL PROLIFERATION

Abstract

This study investigated the preventive effect of diclof enac sodium (DF) on bovine lens epithelial cell (LEC) proliferation. At 24 hr after DF treatment, LEC cell proliferation was determined by the XTT (2, 3-bis [2-Methoxy-4-nitro-5-sulfophenyl] -2H-tetrazolium-5-carboxanilide) test. The contents of b-FGF and TGF-β2 in LEC were measured with an immunoassay kit. The intracellulor free calcium concentration ( [Ca²⁺]i) was determined by conf orcal laser microscope image analysis with fluo-4/AM. LEC cell proliferation decreased with DF treatment. DF had no obvious effect on b-FGF and TGF-β 2 production. [Ca²⁺] i increased with 500pM DF treatment. These data suggest that the preventive effect of DF on LEC cell proliferation might be caused by an increase of [Ca²⁺] i, rather than b-FGF and TGF-β2 production in LEC.
