    Chemotherapy against hormone refractory prostate carcinoma has been investigated. Although occasional responses have been seen, prostate cancer still which shows low responses to chemotherapy. One of the factors which makes cancer cells acquire resistance is heat shock protein (HSP) while cycloheximide (CHX) is known as a substance which inhibits HSP function. Thus, we hypothesized that administration of CHX prior to heat treatment could raise effectiveness an against anticancer reagent. On MTT assay in chemotherapy revealed that heat treatment inhibited efficacy of cisplatinum (CDDP) and THP-adriamycin ( THP) . In addition elevated HSP expression was detected with heat treatment by western blot analysis. From the results, it could be said that HSP inhibited efficacy of CDDP and THP. Subsequently, CHX was administrated prior to the anticancer reagent. The HSP expression was inhibited and CHX made warm the chemothermotherapy more effective. We conclude that CHX heightens the effectiveness of chemotherapy.

    Remodelling of the airways involves a thickening of the reticular lamina of the basement membrane and proliferation of the smooth muscle. IGF-1 stimulates proliferation of a variety of mesenchymal cell types and collagen synthesis. We hypothesis that an increase in IGF-1 progresses airway remodelling. We investigated whether the levels of IGF-1 in 15 mild to moderate asthmatics were higher than in five normal controls. In addition, we examined whether the levels of IGF-1 correlated with FEV 1% and bronchial responsiveness. The IGF-1mRNA expression showed no significant difference between asthmatics and normal controls in bronchial biopsies, but in BALF asthmatics the IGF-1mRNA expression was higher than in normal controls. We observed a significant increase of IGF-1 protein levels (p<0.05), eosinophils (p<0.05), total cells in BALF (p<0.05) in the asthmatics compared with normal controls. This was accompanied by a significant increase in the thickness of the reticular lamina of the basement membrane (p<0.05) . Significant correlation was found between the levels of IGF-1 and eosinophils (%) in BALF (Rs=0.45, p=0.04), and the thickness of the reticular lamina of the basement membrane (Rs=0.56, p=0.01) . These results suggest that an increase in IGF-1 progresses the thickness of the reticular lamina of the basement membrane and an increase in IGF-1 is correlated with eosinophil increase, and therefore, may progress remodelling of the airways.

    This study investigated the preventive effect of diclof enac sodium (DF) on bovine lens epithelial cell (LEC) proliferation. At 24 hr after DF treatment, LEC cell proliferation was determined by the XTT (2, 3-bis [2-Methoxy-4-nitro-5-sulfophenyl] -2H-tetrazolium-5-carboxanilide) test. The contents of b-FGF and TGF-β2 in LEC were measured with an immunoassay kit. The intracellulor free calcium concentration ( [Ca2+]i) was determined by conf orcal laser microscope image analysis with fluo-4/AM. LEC cell proliferation decreased with DF treatment. DF had no obvious effect on b-FGF and TGF-β 2 production. [Ca2+] i increased with 500pM DF treatment. These data suggest that the preventive effect of DF on LEC cell proliferation might be caused by an increase of [Ca2+] i, rather than b-FGF and TGF-β2 production in LEC.

    We report a case of spastic hemiplegia with excessive synergy of the left lower limb, for which a sciatic nerve block was effective. The patient was a 57-year-old female, who had subarachnoid hemorrhage followed by cerebral infarction, and was admitted to our hospital for the purpose of rehabilitation five months after the onset. Left unilateral spatial neglect and left hemiplegia were observed. Synergy from extreme spasticity was also observed, which hindered the extention of the left lower limb. As neither physical therapy nor pharmacological approach produced any effect, a sciatic nerve block by a local anesthetic (1 % lidocaine hydrochloride 10 ml) was tried once or twice a week for a total of 10 times. Spasticity of the hamstrings decreased gradually and the knee extensor muscles started to contract. The power and the movement of the lower limb improved and the subject began to walk again. Improvement of the left lower limb was because of the following: . The nerve block temporarily relaxed the knee extensor muscles and removed reciprocal inhibition. The experience of muscle contraction under such a condition expanded the possibility of lower limb movement. On the other hand, contraction of the extensor muscles produced reciprocal inhibition against the hamstrings, decreasing the spasticity. Thus, the subject was able to walk.
