Gastrointestinal stromal tumours (GISTs) are the most common mesenchymal tumours of the gastrointestinal tract. Some of GISTs lacking mutations in KIT or PDGFRA harbor the non-sense mutation of succinate dehydrogenase (SDH) and the carcinogenesis may be driven by the accumulation of succinate as one of the results of the SDH mutation. Therefore, evaluating the intra- and extra-cellular concentration of succinate is of importance to understand the carcinogenetic process of SDH-deficient GISTs.
Here we established the assay system for succinate concentration. Using Succinyl-CoA Synthase coupled assay, the absorption rate for NTB (λ=412nm) is measured, which indicates the succinate concentration. We evaluated the intra- and extra-cellular succinate concentration of DLD-1 cells under normoxic and hypoxic conditions. Under hypoxic conditions, the cells secreted more succinate than they did under normoxic conditions, suggesting the possibility that hypoxic conditions induce the cells to produce and to secrete succinate, which may promote the carcinogenesis.
