Cell spreading is important for many physiological/pathological circumstances including embryonic development,cancer metastasis and immune cell interactions.A new method for high-throughput study of cell spreading dynamics is devised by multi-step microscopy projection photolithography based on a cell-friendly photoresist.By releasing large number of rounded cells in single cell arrays and monitoring their spreading dynamics by interference reflection microscopy,large number of cell spreading data can be acquired by a single experiment.With this novel experimental setting,we systematically examined roles of various cytoskeleton/motor proteins on cell spreading by using pharmacological inhibitors.
