We report our efforts on evaluating the RNA extraction efficiency via microfluidic ITP (isotachophoresis). We have developed a theoretical model based on second-order kinetics and validated it with experimental results to explore the effect of the key ITP parameters, including target initial concentration, forward and reverse reaction constants, the maximum surface adsorption, ITP zone velocity and electric field. We also report that the polydimethylsiloxane microchannels are compatible with ITP-aided RNA extraction and yields 87.2 ± 9.4% (S. E.) extraction efficiency at single cell level.
