Red blood cells are of biconcave disc shape with high deformability, and water permeates interior through erythrocyte membranes due to the osmotic pressure difference in a low concentration solution (hypotonic solution), and then the erythrocytes expand and leads to hemolysis. In this study, we applied hypotonic solution to erythrocytes and tried to calculate elongation elastic modulus of erythrocyte membranes. After washing the blood by centrifugation, only red blood cells were taken out. For ease of observation, a red blood cell sample was prepared by diluting with MAP solution. NaCl solution of 0.50 wt% concentration was applied to the red blood cell sample dropped on the slide glass and the state of expansion was observed and photographed. From the experimental results obtained, the elongation elastic modulus of erythrocyte membrane was calculated. The value was larger than the reported value. The cause may be thought that the outflow of ions was not considered. Therefore, in the future we will use Hodgkin-Huxley model to examine ion spill.
