Abstract

Pinpoint photooxidative cell membrane perforation method enables mass cell processing with a nano-needle array-shaped perforator formed by nanoimprint process. Hence the perforator still needs improvements for various points, we report the trials on (1) cell damage reduction through the process with focused orientation of photosensitizer, within the perforator; (2) sponge-like perforator to hold materials for intracellular delivery through the process; (3) surface cleansing to recover the perforation function after the usage. As the results, (2) The sponge-like perforator caused negative cell absorption to the surface but other trials achieved positive results; (1) the focused orientation perforator shows enough performance and reduces the cost of its fabrication process; (3) the cleansing process which employs enzymatic treatment plus ultrasonification recovers the perforation function. Finally we tried patterning perforation process by light irradiation area control and the results shows reasonable difference.
