Abstract
We have developed an Arabidopsis inorganic phosphate transporter (PHT1) promoter as a root specific expression promoter applicable to plant recombinant DNA technologies such as phytoremediation. The PHT1 promoter was constructed as fusion of a gene for GUS, and the activity of this promoter was assayed in Arabidopsis and rice plant. As a result, the GUS activity was detected strongly in root, and a negative response for the phosphate concentration was detected in both Arabidopsis and rice. The GUS activity in the organ except for the root was low.
It is indicated that the PHT1 promoter functions via the expression mechanism which the plant commonly has, because it actives not only in a dicot (Arabidopsis) but in a monocot (rice). Therefore, we intend to analyze whether the PHT1 promoter functions in Eucalyptus and poplar which are the practical plants at present.
