Citrate possesses an ability to chelate Al3+ and secretion of citrate from the root apex plays an important role in Al resistance in some plant species. The secretion of citrate is proposed to be mediated by an anion channel on the plasma membrane. In the present study, identification of Al-induced membrane-bound proteins was attempted in rye. Rye secrets both malate and citrate in response to Al and the secretion of citrate was delayed for several hours after the exposure to Al. Membrane proteins were extracted from the root apices (0-5 mm) exposed to Al or not for 12h and subjected to 2D-PAGE analysis. By comparing protein profile between different treatments, 26 Al-induced proteins were detected. Some candidate proteins were then subjected to LC-MS/MS analysis and 16 proteins were hit in the database. The relationship between these proteins and the citrate secretion is now under investigation.
