Abstract
Abscisic acid (ABA) is an important phytohormone that regulates many aspects of plant growth and development. To identify components of the ABA signaling pathway, many screens for mutants have been carried out.
abi5-1, one of the ABA insensitive mutants, can germinate in the presence of ABA. ABI5 encodes a bZIP transcription factor that controls the expression of ABA responsive genes in seed development and germination. Recent studies showed that ABI5 activity is controlled at the protein stability level in ABA dependent manner. To obtain new ABA-related mutants, we constructed a screening method based on the fluorescence of ABI5-GFP or GFP-ABI5 fusion proteins. To date, we verified the expression of those GFP fusion proteins in transgenic plants that reflect ABI5 behavior. And we raised ABI5 antiserum to analyze ABI5 behavior in detail. We report here the constructing process of this monitoring system.
