Abstract
To improve the wood quality, we have been analyzing the gene expression profiles in Eucalyptus xylem. In current study, we are characterizing several transcription factor genes and promoter regions. Fluorescence Correlation Spectroscopy (FCS) using MF20 (Olympus) can analyze the interactions of molecules easily and quickly. Using FCS, interacting molecules is detected as the diffusion time changes.
Firstly, we analyzed interaction between Arabidopsis DREB1A transcription factor and rd29A promoter with MF20. DREB1A binds the DRE motif in rd29A promoter region. As the results, we detected the diffusion time changes showing the DRE-DREB1A interaction. Therefore, FCS is an efficient method to detect interactions between a promoter and a transcription factor. Currently, we are performing the comprehensive analysis of interactions between promoters and transcription factors derived from Eucalyptus xylem using FCS.
