Abstract
We transformed Eucalyptus camaldulensis with a salt-tolerant gene, coline oxidase by the MAT vector system. The regenerated shoots were selected by rooting on a medium containing 200 mM NaCl. Consequently, three lines of the transgenic plants were used for further analysis. In order to release the transgenic plants on the environment, we evaluated the following characteristics compared between the transgenic and wild-type plants at two stages, a growth chamber and a semi-closed greenhouse (1) growth characteristics (2) phenotypic changes (3) expression level of transgene, (4) kinds of chemical substances produced by plants (5) sandwich test of lettuce seed-budding using dry leaves, (6) microorganism flora in soil, and (7) the remaining test of Agrobacterium on plants. In the series of experiments, we concluded that the cultivation and experiments were safely carried out throughout these two stages. At the third stage, these clonal transgenic Eucalyptus plants are growing in the isolated field.
