Abstract
Thellungiella halophilais a model halophyte system with a small plant size, short life cycle, a small genome and an efficient transformation, and can grow in 500 mM NaCl medium. Its genes have a high sequence identity (90-95% at cDNA level) to genes of its close relative Arabidopsis thaliana. Furthermore, T.halophilashows not only high salinity stress tolerance but chilling, freezing and heat stress tolerances, suggesting that T.halophilais a good genomic resource. We constructed a full-length cDNA library of T.halophiladerived from various tissues in addition to whole plants subjected to environmental stress treatments by high salinity, chilling, freezing and ABA, and obtained 20000 clones. We sequenced all clones from each of the forward and reverse direction. Finally, we obtained 9565 individual clones. The content of full-length cDNAs in this library is 88%, which is almost equivalent to the reported values given for Arabidopsis and rice.
